{"id":3310,"date":"2017-12-11T15:25:38","date_gmt":"2017-12-11T14:25:38","guid":{"rendered":"https:\/\/www.toulouse-white-biotechnology.com\/insights\/actualites\/actualite\/high-throughput-strain-engineering-workstation\/"},"modified":"2026-03-16T13:49:47","modified_gmt":"2026-03-16T12:49:47","slug":"high-throughput-strain-engineering-workstation","status":"publish","type":"post","link":"https:\/\/www.toulouse-white-biotechnology.com\/en\/insights\/blog\/news\/high-throughput-strain-engineering-workstation\/","title":{"rendered":"High throughput strain engineering workstation"},"content":{"rendered":"<strong><img loading=\"lazy\" decoding=\"async\" class=\"alignright size-full wp-image-7043\" src=\"https:\/\/www.toulouse-white-biotechnology.com\/app\/uploads\/2017\/12\/equipement-2.jpg\" alt=\"\" width=\"289\" height=\"284\">TWB high throughput strain engineering laboratory developed an original and fully integrated robotic system dedicated to automated strain engineering processes.<\/strong>\n\n<p>The workstation includes an<\/p>\n\n<strong> automated liquid handler<\/strong>\n\n<p>(StarPlus Hamilton Robotics) and a<\/p>\n\n<strong>built-in extension<\/strong>\n\n<p>.\r\nThe liquid handler is fitted out with two pipetting tools:<\/p>\n\n<ul class=\"wp-block-list\">\r\n \t<li>an eight independent channels arm<\/li>\r\n \t<li>a 96 channel pipetting head with a pipetting range of 1 to 1000 \u00b5L.<\/li>\r\n<\/ul>\n\n<p>The carriers and accessorizes on the deck are adapted to molecular biology workflows (cooling and heating shakers, vacuum stations).\r\nThe built-in includes:<\/p>\n\n<ul class=\"wp-block-list\">\r\n \t<li>stackers for microplates,<\/li>\r\n \t<li>an automated incubator for incoming and outgoing biological samples (Thermoscientific),<\/li>\r\n \t<li>a multimode plate reader (Safas),<\/li>\r\n \t<li>a microplate centrifuge (Agilent),<\/li>\r\n \t<li>a microplate dispenser (Biotek),<\/li>\r\n \t<li>a sealer (Fortitude),<\/li>\r\n \t<li>eight independent microbiological incubators (Inheco).<\/li>\r\n<\/ul>\n\n<p>A<\/p>\n\n<strong>robotic arm<\/strong>\n\n<p>moves the plates from the built-in to the liquid handler and to the various integrated devices. A<\/p>\n\n<strong> laminar flow<\/strong>\n\n<p>protects the whole workstation (HEPA filter hood).<\/p>\n\n<strong>The system is devised to be modular, with a range of unit methods that we assemble according to the project, to build a new and original workflow.<\/strong>\n\n<p>Using this robotic tool, we develop automated unit methods for<\/p>\n\n<strong> microbial strain engineering and synthetic biology<\/strong>\n\n<p>: several DNA assembly methods, genome editing using CRISPR-CAS9, automated microbial strain transformation for<\/p>\n\n<em> E. coli, S. cerevisiae<\/em>\n\n<p>and<\/p>\n\n<em>Y. lipolytica<\/em>\n\n<p>. In addition, we develop<\/p>\n\n<strong>methods to screen the constructs<\/strong>\n\n<p>we obtain: genotyping, enzymatic tests, automated fluorescence and bioluminescence measurements, growth properties, protein expression optimization by expression screening and affinity purification of tagged recombinant proteins. The tools combine the flexibility and the throughput we need to provide customized workflows for various microorganisms\u2019 backgrounds.
