{"id":3823,"date":"2016-03-30T22:16:09","date_gmt":"2016-03-30T20:16:09","guid":{"rendered":"https:\/\/www.toulouse-white-biotechnology.com\/insights\/actualites\/actualite\/the-nucleic-acid-preparation-automated-system\/"},"modified":"2026-03-16T13:52:53","modified_gmt":"2026-03-16T12:52:53","slug":"the-nucleic-acid-preparation-automated-system","status":"publish","type":"post","link":"https:\/\/www.toulouse-white-biotechnology.com\/en\/insights\/blog\/news\/the-nucleic-acid-preparation-automated-system\/","title":{"rendered":"The nucleic acid preparation automated system\u00a0"},"content":{"rendered":"<strong><img loading=\"lazy\" decoding=\"async\" class=\"alignright size-medium wp-image-4679\" src=\"https:\/\/www.toulouse-white-biotechnology.com\/app\/uploads\/2016\/03\/capture-decran-2016-03-30-a-21.53.22-300x197-2.png\" alt=\"Capture d\u2019\u00e9cran 2016-03-30 \u00e0 21.53.22\" width=\"300\" height=\"197\">The robot (Hamilton Robotics) installed on TWB\u2019s high-throughput\u00a0microbial strain engineering platform enables the automated preparation of nucleic acids.\u00a0<\/strong>\n\n<p>The robotic liquid handling platform performs, in a<\/p>\n\n<strong>96-well microplate format, extractions and purifications of nucleic acids from different types of micro-organisms<\/strong>\n\n<p>: plasmid DNA, genomic DNA, DNA fragment purification and RNA\u00a0purification.\r\n\r\nThe system uses both to use<\/p>\n\n<strong>silica gel adsorption techniques and magnetic bead-based immobilization techniques<\/strong>\n\n<p>.\r\n\r\nThe manipulation zone is protected by laminar flow (air supply ceiling\u00a0equipped with a HEPA filter).\r\n\r\nThe robotic platform is equipped with two pipetting tools, one pipetting arm with\u00a08 independent channels and a 96-channel pipetting head\u00a0both pipetting a volume range of<\/p>\n\n<strong>1\u00b5L to 1mL<\/strong>\n\n<p>. The configuration of the robot\u2019s deck is designed to<\/p>\n\n<strong>parallelize two 96-well plates<\/strong>\n\n<p>: two vacuum stations (filtration on filter plates), two magnetic stands (extractions by immobilization on magnetic beads), two agitators for microplates and two thermostated modules (4-99\u00b0C).<\/p>\n\n<figure class=\"wp-block-image\"><img loading=\"lazy\" decoding=\"async\" class=\"aligncenter size-full wp-image-4678\" src=\"https:\/\/www.toulouse-white-biotechnology.com\/app\/uploads\/2016\/03\/capture-decran-2016-03-30-a-21.52.20-1.png\" alt=\"Capture d\u2019\u00e9cran 2016-03-30 \u00e0 21.52.20\" width=\"758\" height=\"503\"><\/figure>\n\n<p>Several<\/p>\n\n<strong>additional pieces of equipment<\/strong>\n\n<p>are used for the<\/p>\n\n<strong>quality control of purified nucleic acids<\/strong>\n\n<p>:<\/p>\n\n<ul class=\"wp-block-list\">\r\n\t<li>A UV-visible micro-volume multi-sample Nanodrop 8000 spectrophotometer for measuring\u00a0concentrations by measure of\u00a0absorbance<\/li>\r\n\t<li>A 96-well pre-cast agarose gel electrophoresis system for the quality control of supercoiled DNA<\/li>\r\n\t<li>Two systems of capillary electrophoresis with average throughput (Bioanalyzer Agilent) and high throughput (GXII Perkin Elmer) for the quality control of linear DNA and quality assessment of RNA by RIN (RNA Integrity Number).<\/li>\r\n<\/ul>\n\n<p>This type of robot is commonly used for several types of applications and projects:<\/p>\n\n<ul class=\"wp-block-list\">\r\n\t<li>Synthetic biology, genetic and metabolic engineering, strain engineering<\/li>\r\n\t<li>Enzyme evolution<\/li>\r\n\t<li>Protein expression<\/li>\r\n\t<li>DNA libraries construction.<\/li>\r\n<\/ul>\n\n<strong>The nucleic acid automated purification system is part of larger robotics installations. It complements in particular an automated strain engineering station currently being set up.<\/strong>\n\n<p>The full implementation of the standard protocols for\u00a0nucleic acid purification is scheduled for<\/p>\n\n<strong>June 2016<\/strong>\n\n<p>.
